Beginning with FDA news, the administration has accepted for review the biologics license application (BLA) for zelecimentrostudirsen (z-rostudirsen, also known as DYNE-251) for Duchenne muscular dystrophy (DMD) amenable to exon 51 skipping.¹The agency granted Priority Review and set a PDUFA target action date of January 21, 2027. Dyne Therapeutics continues to expect a potential U.S. launch in the first quarter of 2027, assuming approval on the anticipatedtimeline.¹
John Cox, president and chief executive officer of Dyne, said in a statement¹, "This milestone represents significant progress toward our goal of delivering functional improvement for those living with DMD amenable to exon 51 skipping. With z-rostudirsen, we set out to advance the treatment paradigm in DMD by combining a robust increase in near-full length dystrophin with broad delivery to relevant tissues."
The submission seeks Accelerated Approval based on dystrophin as a surrogate endpoint, a pathway already established for other exon-skipping therapies in DMD.
In other news, the Alzheimer's Association announced the launch of the PROTECT-Cog Study (Prevention of Risk for cognitive decline through Combined Therapy) at the 2026 Alzheimer's Association International Conference (AAIC) in London. The global trial will evaluate whether combining a multidomain lifestyle intervention with a metabolism-targeting drug, such as a GLP-1 receptor agonist, further reduces cognitive decline risk beyond what lifestyle intervention alone has shown.2
"PROTECT-Cog builds directly on what we learned from U.S. POINTER and takes the next critical step in prevention science," Maria C. Carrillo, PhD, chief science officer and medical affairs lead at the Alzheimer's Association and principal investigator of the study, said in a statement.2 "By testing a combined approach that targets both lifestyle and biology, we have the opportunity to better understand how to meaningfully reduce the risk of cognitive decline before symptoms begin."
The study extends the Association's existing prevention infrastructure, built through U.S. POINTER, LatAm FINGERS, and the World-Wide FINGERS Network. "The Alzheimer's Association is uniquely positioned to lead this next generation of dementia prevention research," Heather M. Snyder, PhD, senior vice president of medical and scientific relations at the Association and staff lead on the project, said in a statement.2
